A general rather than site- & function-specific role for protein modification by ADP-ribosylation?

We find that PARP7 is stabilised following proteotoxic stress and ADP-ribosylates proteins to enable their degradation. This occurs in a ubiquitin-dependent manner.
PARP7 is a proteotoxic stress sensor that labels proteins for degradation | The EMBO Journal
imageimageADP-ribosylation is thought to occur on specific sites to affect specific characteristics of target proteins. Here, it is found to play a more promiscuous role, with PARP7 labelling proteins for efficient degradation during proteotoxic ...

Q for the hive-mind 🧠: What transfection reagents do people like for big constructs these days? (I am thinking #HSV encoding BACs, ~170 kb). We've recently had poor performance with Trans-IT LT1 and Lipofectamine 2000, our two go-tos. Target cells are U2OS. Suggestions or RTs greatly appreciated 🙏

Reminder: Nobel-prize winning PCR (1983), used in basically all genetic tech today, was only possible because of extremophile bacterium discovered in 1964 in Yellowstone funded by a small ~$80k NSF grant with no obvious application at the time. #science 🧪
