I spend a bit of time talking to folk with plasma cohorts about MS vs affinity approaches for discovery. Thought it was worth a thread.

Most love the marketing. 5,000 proteins? 10,000 proteins? OLink was used in UK Biobank? Wow, mass spec is sooooo last year. Except life is not that simple.
Somalogic, Olink, and Alamar are casting doubt on each other’s data, proudly calling out lack of correlation.

I know why they do this but in the long run as it casts doubt on affinity approaches as a whole.

We will think lots of data is generated that is trash and won’t know which

One of the good perils about #TeamMassSpec being active again is, it takes a while to go through all the posts and links..

The side-by-side comparison of volcano plots showing how different the protein differential expression analysis is based on the normalization strategy employed is awesome..
2/ iPSCs and ESCs express nearly the same proteins, yet at different levels. Notably, iPSCs express >4,000 proteins 1.5x higher, with higher overall protein content! This calls for careful normalisation —size or content differences could mask major findings if you simply median normalise. ⚖️

Lots of lovely data for folks interested in #Immunology and #Neurodegeneration!

It has proteome data for ex vivo AND in vitro mouse AND human microglia AND in vitro h-ESC microglia that were xenografted!

📢Intro post. I'm a postdoc at the Centre for Inflammation Research in Edinburgh.

My focus is on using mini-bulk and single cell proteomics to functionally characterise human neutrophils (and other leukocytes) in health and inflammatory disease. My content is a mix of proteomics and immunology. 🧪
