We found diverse pathways to resistance led to a shared vulnerability to CDK12/13 inhibitors, which exacerbated mitotic arrest through loss of CDK12-dependent TP73 expression. Combining CDK12/13 and KRAS inhibitors up front prevented the onset of drug resistance!

We analyzed 9,230 lung adenocarcinoma (LUAD) tumors to assess the mutational context and oncogenic potential of KRAS G12A and G12V and G12C and G12D variants. Their cancer effect sizes were similar—large compared to variants at other sites of KRAS.
