Siglec-9 is an inhibitory receptor that tones down immune responses, in this case, blocking antigen presentation by the cells of the immune system. If this mutation is reverted to the pre-Omicron version antibody responses to Omicron increased by about 10-fold. Furthermore, when the mutation was...

Upon engagement with Siglec-9, antigen-presenting cells like macrophages have reduced function, and in mice, introducing this mutation into the delta spike protein (S375F) resulted in a ~10-fold drop in antibody titers. And reverting it with Omicron resulted in an ~10-fold increase in titers.

When glycan recognition is linked to immune response, you need fine tuning! 😎

Here ⬇️ we show how human Siglecs act as molecular precision tools, recognising the glycan and also the biological environment (or context) these glycans are found in. A 1/5 #glycotime🧵 ⬇️

In our contribution, @silviadandrea.bsky.social and I looked at the binding of GT1b (Siglec-10 fav buddy), which is polysialylated and has a very fancy structure 😍 MD simulations show how *multiple Arg* contribute to the recognition and binding, providing an atomistic perspective to experiment ⬇️ 2/3
