We used SeqHub's CoSearch to explore CRISPR-associated transposon (CAST) systems and found interesting patterns. By searching for TnsB + Cas12 together (Liu et al., 2025), we compared genomic context and architecture across organisms, making it easier to reason about CAST subtypes.

Brand new preprint from my lab, showing that TnpB, the ancestor of Cas12, acts as a gene drive in plasmids! And it turns out in conjugative plasmids that it acts as a primitive anti-self defense system, providing a potential link between its transposon effect and becoming CRISPR!
What is the best strategy to win any contest?

Recently, my friend @fernpizza.bsky.social showed how plasmids compete intracellularly (check out his paper published in Science today!). With @baym.lol, we now know they can fight.

5/8 We engineered this transition: by artificially splitting ISDra2 TnpB’s reRNA into tracrRNA and crRNA, we converted it into a functional CRISPR-like system. This confirms RNA splitting was a key step in Cas12 emergence.

2/8 Phylogenetic mining revealed several young Cas12 clades we call “TranCs”, i.e., nascent CRISPR systems emerging from distinct TnpB lineages. These represent multiple independent domestication events from transposons.

80% is so high tho! It's kind of trivial to find <80% variants of known functional Cas. So if <80% fair game is really true, surely crispr parents would have been meaningless from the start? Cas12, CasX, CasY, CasPhi... these are all basically different folds so I get those as unique IP
