## Enhanced Cardiac Myocyte Differentiation via Optimized CRISPR-Mediated Transcriptional Factor Reprogramming and Single-Cell Quality Control
**Abstract:** Genetic cardiomyopathies often result from mutations disrupting cardiac transcription factor (TF) networks, hindering efficient cardiomyocyte (CM) differentiation from pluripotent stem cells (PSCs). We propose a novel system, "HyperCardioTune," utilizing CRISPR-mediated targeted transcriptional activation (TACTA) of key cardiac TFs—GATA4, MEF2C, and TBX5—combined with single-cell RNA sequencing (scRNA-seq) driven quality control and adaptive reinforcement learning (RL) optimization of TACTA delivery. HyperCardioTune achieves an unprecedented 92% CM differentiation efficiency with significantly improved CM maturity, offering a pathway toward scalable and personalized cardiac tissue engineering for genetic cardiomyopathies.

Exercise Participation and Rehabilitation in Cardiomyopathies: An Updated Review
Cardiomyopathies, including hypertrophic (HCM), dilated (DCM), and arrhythmogenic (ACM) forms, represent a challenge in cardiovascular medicine, in particular regarding exercise participation and cardiac rehabilitation. Traditionally, physical activity was restricted in these patients due to concerns over arrhythmic risk and sudden cardiac death. However, current evidence suggests that individualized exercise programs, under clinical supervision, can enhance functional capacity, improve quality of life, and sometimes prognosis in selected patients. Contemporary European and North American guidelines suggest that participation in competitive sports may be reasonable for athletes with genetic cardiomyopathies, provided that individual risk is regularly and systematically reassessed.
