Pts w/ MRD+ MM undergoing autoHCT (MEL->ASCT->ARM coated NK cells-> IL-2). No DLTs seen at 5-10 million cells/kg. NK cells expand/persist for 60d, are functional, have cytotoxic gene signature enriched for IFNg, infiltrate BM. Too early to assess clinical impact vs SOC ASCT (2/2) #ASH25

and make 50 ng/ml, and upon stimulation they make 100 ng, that is a 2-fold increase. But, which cells make more IFNg? That is 100 ng vs 4 ng. Like with proportions, be very carefull what you are actually looking at, and what conclusion it does or not, support.

Before looking at the data, it is important to uderstand what "fold-change" is. Fold-change shows relative change, not actual impact! If your cells make little IFNg (1 ng/ml), but upon stimulation a little more (4 ng), that is a 4-fold increase. If you cells are more active

The strongest transcriptional signal in ICI-AIN is a IFNg response signature. Interestingly, the IFNg is mostly produced by CD8 T cells that have a more exhausted/tissue-resident phenotype.

scRNA-seq & flow cytometry showed AcAc increases tumor MAIT cells and enhances their expression of cytotoxic effectors like IFNg and granzyme B.
Crucially, the anti-tumor effect was lost in mice lacking MAIT cells, showing they are essential for the anti-tumor effects. (4/10)

More specifically, very recent infection displayed reduced IFNg production and more geared towards IL2 and TNF while TB disease was associated with dual production of IFNg and TNF.

those people infected (red) have many of these T cells (depending on the HLA). In addition, you can find some (blue) in the memory (IFNg+) population of people who have not knowingly been infected.
The frequency is low, since they were not exposed.

A major gene that links inflammation to leakiness is MYLK (myosin light chain kinase).

It gets turned on by TNF and IFNG.

High MYLK → cells contract and pull junctions apart → leak increases.

It’s a key driver in many leaky gut conditions.
