I am excited to share our new preprint on the CAGE complex, a mysterious hollow protein complex that I first saw years ago while surveying Tetrahymena ciliary lysate www.biorxiv.org/content/10.1... #cilia #protistsonsky 🧬🧪

Our preprint, in collaboration with Rino Rappuoli & Emanuele Andreano, is out! Co-led by colleagues at TLS, my grad student Ling Zhou @lingzhou.bsky.social, and postdoc Emily Rundlet @cryoemily.bsky.social, we identified OPG153 as a poxvirus bnAb target via B cell isolation & AlphaFold3 prediction.

Latest preprint from the lab, many years in the making!

By combining #cryoEM with #AlphaFold3 modelling, we propose that norovirus NS3 forms a transmembrane RNA translocase.

This could have big implications for our understanding of viral replication & assembly (🧵)

Grateful to be a part of such a fun project led by my labmate @luqiang.bsky.social!
Our latest preprint is now available doi.org/10.1101/2025...! Led by my postdoc, Luqiang Guo @luqiang.bsky.social, we determined a ~2.4 A resolution structure of the hantavirus glycoprotein tetramer on the surface of VLPs. Also determined structures of dimers of tetramers and Fab-bound tetramers.

Check out our latest study, led by grad student Ryan McCool @rmccool.bsky.social‬, in collaboration with Vaccine Company. We were able to stabilize EBV gB in the prefusion conformation, determine its cryo-EM structure to 3.1 A, and assess its immunogenicity in mice. www.biorxiv.org/cgi/content/...

Happy to finally see this study published, a great collaboration between academic, government and industry labs! We show that CCHFV GP38 can induce vascular leak and that this can be prevented by protective but not virus-neutralizing antibodies! www.science.org/doi/10.1126/...
Antibodies targeting Crimean-Congo hemorrhagic fever virus GP38 limit vascular leak and viral spread

I’m super excited to see this work out from my lab/PhD-cohort-mate @elizabeth-mcfadden.bsky.social. By engineering and structurally analyzing a soluble gp38-Gn-Gc complex, she develops new insights for CCHFV and other type II viral fusion proteins alike. Definitely worth the read!

Congratulations Elizabeth! Great collaborative effort with our colleagues Kartik Chandran, Andrew Herbert, Stephanie Monticelli and more!
This is my first, first-author paper of my PhD and is a reminder of my growth over the last several years and the excellent training and mentorship I've received from
@mclellanlab.bsky.social. Thanks to all who contributed to this work!

Engineering and structures of Crimean-Congo hemorrhagic fever virus glycoprotein complexes
Crimean-Congo hemorrhagic fever virus is a widespread tickborne virus that infects humans. Engineering-based structural studies of the viral glycoprotein complexes GP38-Gn and GP38-Gn-Gc provide insights to their function in the viral life cycle. GP38-Gn-Gc confers partial protection against lethal viral challenge in mice, showing promise as a vaccine antigen for further development.
