Phase I trial shows promise for hES cell-derived dopaminergic neurons in Parkinson's. NCT04802733 offers hope for tackling disease progression. PMID:40240592, Nature 2025, @Nature https://doi.org/10.1038/s41586-025-08845-y #Medsky #Pharmsky #RNA #ASHG #ESHG 🧪
Phase I trial of hES cell-derived dopaminergic neurons for Parkinson’s disease | Nature
Parkinson’s disease is a progressive neurodegenerative condition with a considerable health and economic burden1. It is characterized by the loss of midbrain dopaminergic neurons and a diminished response to symptomatic medical or surgical therapy as the disease progresses2. Cell therapy aims to replenish lost dopaminergic neurons and their striatal projections by intrastriatal grafting. Here, we report the results of an open-label phase I clinical trial (NCT04802733) of an investigational cryopreserved, off-the-shelf dopaminergic neuron progenitor cell product (bemdaneprocel) derived from human embryonic stem (hES) cells and grafted bilaterally into the putamen of patients with Parkinson’s disease. Twelve patients were enrolled sequentially in two cohorts—a low-dose (0.9 million cells, n = 5) and a high-dose (2.7 million cells, n = 7) cohort—and all of the participants received one year of immunosuppression. The trial achieved its primary objectives of safety and tolerability one year

Neurons transplanted into a longer-living organism will continue living up to the end of that organisms lifespan:

This gives hope for human lifespan limited only to how long we can keep the brain alive and healthy, even outside the body.

When you think of the long and gloomy history of man, you will find more hideous crimes have been committed in the name of obedience than have ever been committed in the name of rebellion. (Snow, 1961, p. 24)

Upon infection, the C. elegans worm reshuffles the roles of brain cells and flips the functions of some of the chemicals it uses to regulate behavior.

Never forget to celebrate Rosalind Franklin’s work and legacy.
When F. Crick, J. Watson and M. Wilkins were awarded the Nobel Prize for their work on DNA, no reference was made to Franklin’s significant contribution to this discovery.
