5️⃣ Lastly, by integrating the atlas data with in vitro models of induced EC senescence, we uncovered a core set of genes linked to CDKN1A/p21 expression. We validated this across models, tissues and species, to further our understanding of senescence regulation in ECs. 7/9

4️⃣ Age-associated ECM remodeling signatures were linked to increased activity of #mechanotransduction regulators YAP/TAZ and TEAD, particularly in skin, lung, and liver. Immunofluorescence confirmed increased nuclear YAP in aged human skin ECs. 6/9

3️⃣ Venous aging is different:
Vein ECs lose expression of multiple angiogenic regulators with age, while increasing genes linked to extracellular matrix (ECM) remodeling and altered glycosylation, hinting at effects on #angiogenesis, barrier function, and immune trafficking. 5/9

2️⃣ Inflammaging is everywhere:
Across tissues, aging ECs showed increased expression of inflammatory signaling, immune cell adhesion markers, and #senescence-associated factors. 4/9

1️⃣ A human vascular “Rosetta Stone”:
We identified conserved molecular markers for each endothelial subtype across tissues. These can be used to pinpoint arterial, venous, capillary, or lymphatic ECs in any healthy human tissue context. 3/9
