We sequenced the anti-RSV-F antibody 131-2a by mass spec and determined its epitope by cryoEM. New insights in postfusion specific binding and antigenic site I. Now published in ACS Infectious Diseases @pubs.acs.org.

The improved (depth of) coverage improves sequencing accuracy and potentially saves a lot of instrument time, because so far we’ve always had to run multiple parallel protease digestions.

This yields way more unique peptides than trypsin or even chymotrypsin. And the peptides are also often longer, which is great for assembly in de novo sequencing of antibodies.

These new archaeal proteases are great for de novo antibody sequencing by mass spec. Great work by @tshamorkina.bsky.social , Laura and @tkadava.bsky.social together with @hecklab.bsky.social.

Lots of the O-glycans are present in N-linked NXS/T sequons. We first noticed this in EBOV GP and since paying attention we see this again and again now also in these coronavirus spikes. Wonder what kind of crosstalk is going on here 🤔

We also find spurious O-glycosylation throughout the spike, with a more dense hotspot in a mucin-like region.

We mapped N- and O-linked glycosylation in these unusual spikes by mass spec. Paints a picture of the glycan shield of this important antigen.
