Perturbing H3K27ac in donor nuclei before reprogramming via p300/CBP inhibition, we observed an improvement of the ON-memory phenotype in cloned embryos, and increased developmental rates. But is H3K27ac on promoters or enhancers in donor cells the culprit for ON-memory in NT embryos?

We then used this data to develop Digital Reprogramming, a machine learning-algorithm that could predict genes that are reprogramming-resistant or susceptible to reprogramming in nuclear transfer. Furthermore, Digital Reprogramming uncovered H3K27ac as a novel candidate barrier to reprogramming.
