Interested in studying #nucleosomes, #chromatin, or #chromosomes at the single molecule level, atomic force microscopy (#AFM) is easy to use technique. Here is a great methods paper to guide you through the whole process. Big thanks to Keir Neuman and Yamini Dalal.

Single-Molecule Studies with AFM and High-Speed AFM: From Nucleosomes to Chromosomes
Atomic force microscopy (AFM) is a single-molecule imaging technique invented in 1985. Development of high-speed AFM (HS-AFM) in 2001 now allows for the direct visualization of the dynamics and kineti...

Our latest work. We identified two structurally distinct CENP-A nucleosomes at the centromere. One is associated with the CCAN and both are important for regulating centromeric transcription.
Centromeres contain two physically distinct CENP-A nucleosomes, one of which associates with the CENP-C complex and regulating CENP-C levels is critical for accessibility of the centromeric chromatin fiber for transcriptional machinery @dpmelters.bsky.social https://buff.ly/40LsByD

Our latest work is now online.
CENP-C is critical in regulating centromere homeostasis by supporting a unique higher-order structure of centromeric chromatin and altering the accessibility of the centromeric chromatin fiber for transcriptional machinery @dpmelters.bsky.social https://buff.ly/40LsByD

The latest work from the Dalal lab. @sweta_sikder and co show how p53, CENP-B, and LSD1 can help rescue CENP-A and CENP-C levels in aged cels.

Centromere inactivation during aging can be rescued in human cells
Sikder et al. uncover how centromere function is depleted during human aging. They report reduced CENP-A/C levels and centromeric transcription in aged cells. Using knockdown approaches, they identify...

I’m proud to have contributed to this beautiful work by Yvonne Fondufe-Mittendorf lab, a project led by now Dr. Wesley Saintilnord and co — Aberrant expression of histone H2B variants reshape chromatin and alter oncogenic gene expression programs www.biorxiv.org/content/10.1...
Aberrant expression of histone H2B variants reshape chromatin and alter oncogenic gene expression programs
Chromatin architecture governs DNA accessibility and gene expression. Thus, any perturbations to chromatin can significantly alter gene expression programs and promote disease. Prior studies demonstra...
