It’s been a hectic couple days in lab but put together so many things. Made (in vitro) and transfected mRNA for the first time, produced Lenti for the first time, have some edited iPSCs and NPCs, incubators full of CHOs expressing proteins.
Just very satisfying when large projects come together.

Going to throw this one out there.

Cloning into a vector to produce some mRNA. The vector sequence itself has the PolyA. Sequenced Plasmid at @plasmidsaurus.bsky.social - showing up as chopped at the poly(A) sequence. Does ONT sequencing struggle with this? Only getting about 35/105 A’s.

amine amino imine amide azide should have consistent pronunciation since they’re all referencing the nitrogen. Instead we have a hodgepodge of syllable emphasis and short/long vowel problems.

Should be either A-mean/A-mean-o/I-mean/A-mide/ A-zide or the emphasis on the second syllable.

1. Imagine we land a space probe on one of Jupiters’ moons, take up a sample of material, and find it is full of organic molecules. How can we tell whether those molecules are just randomly assembled goo or the outcome of some evolutionary process taking place there? 🧪

When I was to hate the detail work, and only wanted to do the broad strokes and dream big. I kind of always just told myself it was easy for other people so I didn’t need to do it. As I grew up I realized that detail work isn’t easy for anyone, and it’s literally my job to do it.

Babysitting iPSCs and pushing them to NPCs for the first time ever. Seems like it’s going well, I see lots of projections. No clue what a rosette actually looks like other than on paper, I guess we’ll find out soon enough.
