Phosphorylation-dependent tuning of mRNA deadenylation rates - Nature Structural & Molecular Biology
Stowell et al. show that the intrinsically disordered region (IDR) of Puf3 binds the Ccr4–Not deadenylase complex using an extended interface. Phosphorylation of the IDR regulates the interaction to t...

A fun collaboration with @rasilab.bsky.social led by Rachael Bakker.

We used an RNA base editor to analyze at scale how substitutions within an RNA or RNA-binding protein affect binding.

Happy to see this work published in RNA, and thanks to the editors for the interview/Meet the First Author Feature! Surprisingly, we found that mutating an RNA binding protein's ability to bind RNA did not remove all biological function. #RNAsky #RNAbiology.

It was great working with Chen Qiu, Traci Hall, and all the other authors on this project! Using a combination of structural, biophysical, and in vivo stem cell analyses, we investigated how an RBP and a partner regulate adjacent binding elements in a critical RNA.

A higher order PUF complex is central to regulation of C. elegans germline stem cells
Nature Communications - PUF proteins, conserved stem cell regulators, maintain germline stem cells with partner proteins in nematodes. Here, authors discover a complex of two FBF-2 PUF proteins and...

I’m excited to share this story. We found that mutating an RNA binding protein’s ability to bind RNA did not remove all biological function, but when combined with a mutation that affects protein interactions, the RBP no longer functioned.

The PUF RNA-binding protein, FBF-2, maintains stem cells without binding to RNA
Like all canonical PUF proteins, C. elegans FBF-2 binds to specific RNAs via tripartite recognition motifs (TRMs). Here we report that an FBF-2 mutant protein that cannot bind to RNA, is nonetheless b...

First Bluesky post, and I'm so excited to share our latest work, online today! We found a protein-protein interaction interface on a Pum/Puf protein mediates binding to a specific subset of RNA targets, and in turn affects only one of its functions in stem cells. 1/2
