Microglia activate SPI1/FLI1 immune programs, while TFDP1 diverges between diseases. Astrocytes engage AP-1 stress pathways; APOE↓ in AD and HIF1A↓ in PiD mark distinct metabolic states. Oligodendrocytes show HES1/ZBTB33 repression shaping myelin & tau-related vulnerability.

TF occupancy shows shared genome-wide TF gains (BHLHE22), AD-specific gain (JDP2), and loss of CTCF.
Fine-mapped SNPs disrupt ADAM10 regulation, and a key gene UBE3A↓ in PiD. PiD shows broad TF down-regulation.

We mapped AD/FTD GWAS (PIP>0.95) to cell-type–specific open chromatin. 50% of AD signals fall in microglial enhancers. GLDN and others emerge as PiD candidates. We constructed SNP→cCRE→gene links and TF occupancy, which is available via scROAD (swaruplab.bio.uci.edu/scROAD/).
