(3/n) Our study reveals that transcription factor combinations use distinct strategies to locate enhancers:
• OSKM factors leverage motif orientation within chromatin loops to target pluripotent enhancers.
• GETM factors use looping junctions to locate trophectoderm enhancers.

(2/n) We identified a previously uncharacterized DNA element adjacent to enhancers, which we’ve termed the "signpost." This element facilitates transcription factor binding near enhancers, enabling precise targeting and activation.

Happy to share our recent collaborative study with the Cedar and Stanger labs, identifying the barrier responsible for incomplete epigenetic reprogramming in both in vitro and in vivo transdifferentiation models.
