By deciphering mechanisms in zebrafish, can we find the missing factors for progenitor maintenance in humans?
This can propose stepping stones towards future female regenerative reproductive medicine.

We discovered ovarian modules in zebrafish and humans - morphogenetic units wherein germline stem cells/early progenitors produce differentiating oocytes in a spatially linear developmental manner, controlled by #Notch signaling.

In species with non-synchronous ovarian development like zebrafish & humans, the production of oocytes from early progenitors is poorly understood. In zebrafish, germline stem cells produce oocytes throughout life, but how they are maintained and produce oocytes is unknown.

I can now share that during the recent Israel-Iran conflict, our main #zebrafish husbandry facility collapsed abruptly, resulting in the loss of ~85% of our fish.

With careful management, we were able to preserve all lines, and are currently operating from backup systems.

By combining genetics, pharmacological manipulations, and rescue experiments, we constructed a novel mTOR-Stat3-Stathmin pathway that orchestrates centrosome regulation and microtubule organization, thereby establishing oocyte polarization competence.

Upstream, we identified mTOR as activator of Stat3.

mTOR is active in oocytes (A), and required ex-vivo for polarity (B) and for Stat3 activation (C).

Stathmin inhibition rescued polarity in mTOR deficient ovaries, confirming that mTOR acts upstream of Stat3-Stathmin (D-E).

Stathmin inhibition on stat3-/- background also rescued microtubule (B) and centrosome (C) organization, strongly supporting our hypothesis and demonstrating that Stathmin acts downstream of Stat3.

If this hypothesis is correct, inhibiting Stathmin in stat3-/- ovaries should prevent microtubule destabilization and rescue polarity.
Indeed, we identified Stathmin in oocytes (A-B), and Stathmin inhibition on stat3-/- background ex-vivo rescued polarity (C).
