The simulations also showed that lipids are scrambled via a credit-card mechanism assisted by presence of polar TRAP residues and membrane thinning. Simulations also provided insights into lipid selectivity, thermodynamics, and kinetics of scrambling.

We next investigated the role of Sec61 translocon together with the heterotetrameric TRAP complex, which stably associates with Sec61. MD simulations demonstrated that both Sec61 gate and a polar groove formed by TRAP beta and delta subunit transmembrane regions are sites of lipid scrambling.

Lipid scrambling assays with fluorescent lipids and dithionite and BSA quenching clearly demonstrated that reconstituted Sec61 translocon had lipid scrambling activity the tested lipids (PC, PS).

Our earlier work revealed the structure of the conserved Sec61/TRAP translocon complex and local membrane membrane thinning around Sec61/TRAP, which led us to speculate whether lipid scrambling might occur in proximity of the translocon.
