For example, we saw ~2000 skipped exon events due to the compound in both cell lines but only ~25% of those were shared. The remaining 75% were unique to either WT or the mutant cells.

Finally, Sierra worked with Hai Dang's group at U. Minnesota to show that splicing factor mutant cancer cells (SRSF2-P95H for aficionados) were sensitive to one of these compounds.

From these she found four that not only caused accumulation of the splicing reporters, but actually impacted splicing of an endogenous transcript in strain-specific ways.

Sierra screened 10s of thousands of compounds and found several hundred that inhibited yeast growth in strain-specific ways.

In this work, Sierra developed a yeast-based screen to look at splicing inhibitors using sensitized yeast strains (that contained "humanized" versions of SF3B1/Hsh155 and Brr2).

We propose that the alpha finger is dynamic in response to the 3'SS: folding and unfolding. Only the folded form allows for docking of the Prp22 c-terminal domain.
