It was a great pleasure to share our work on annelid regeneration at @msarscentre.bsky.social last week and to discuss with the great students and PI. The visits of animal facilities was a blast ! Thank you @lionlchristiaen.bsky.social for the invitation at #sunnyBergen !

In this study, led by former PhD student Loïc Bideau, we showed that Notch regulates neural progenitor specification and is important for axon guidance by elegantly combining HCR expression, functional assays, differential RNA-seq and fine dissection of the neurogenic cascade.

Finally, we showed that ROS inhibition differentially affects tissue identity in Platynereis, suggesting a signal between the ROS-producing ectodermal cells and the ROS-responding meso-endodermal cells that proliferate.

On the other hand, in Nematostella ROS production is required for a second episode of injury-induced apoptosis and for the sustainment of proliferation in later stages, particularly in the body wall epithelia at the amputation site.

ROS inhibition in Platynereis does not affect injury-induced apoptosis but reduces cell proliferation in the regenerating structures at all stages, more strikingly in the meso-endoderm.

ROS are required for regeneration in both models, with inhibition of ROS production blocking the process at early stages.

We investigated more specifically the role of ROS during posterior regeneration in Platynereis (annelid) and oral regeneration in Nematostella (cnidarian). We showed that ROS are produced in both models, but their dynamics rely on distinct enzymatic activities.

But the precise composition of the redox arsenal can vary drastically from species to species, suggesting that evolutionary constraints apply to ROS metabolism functions rather than precise actors.

Thanks to a detailed inventory of the main ROS metabolism actors across metazoans, we established that the vast majority of metazoans possess a core redox kit allowing for the production and detoxification of ROS, and overall regulation of ROS levels.
