This project demonstrates that MIXL1 is directly linked to definitive endoderm generation, similar to its role in mice, and that its expression level could be predictive for cell lines amenable to functional tissue generation, which is a prerequisite for stem cell therapy. (6/7)

These lines were subjected to differentiation on micropatterns. We found that increasing MIXL1 expression levels led to the production of more definitive endoderm cells. (5/7)

The second part consisted of a functional study of MIXL1. We engineered mutant hiPSCs using CRISPR-Cas9 and created an inducible line using CRISPRa methods. (4/7)

Then, by combining scRNA-seq data, we validated our findings on 11 hiPSC lines and extended them to a larger cohort of 125 cell lines from @steglelab.bsky.social (3/7)
