7/8 We believe that systems-level immunomonitoring offers an important complement to tumor genetics and can enable real precision medicine for children with cancer as exemplified below:

6/8 Clonal expanded mem CD8 T cells are transcriptionally similar in children and adults with diverse tumors, but the overall freq. of expanded cells is lower in children, suggesting that initial triggering, not removing the breaks (checkpoint inhib.) is most likely to improve outcomes in children

5/8 Tumors of children are less mutated on average, but there is significant variability within tumor types and among children and mutation rates correlate with CD8+ T cell infiltration in Wilms and Neuroblastomas...

4/8 There are age associated changes in cell composition and system inflammatory responses (ex Neutrophil-to-Lymphocyte ratio) which need to be considered when developing immunotherapies tailored to the immune systems of children of different ages...

3/8 To assess immune variation and determinants of productive immune responses we recruited 191 children with diverse solid tumors, age 0-18yrs at Astrid Lindgren children's hospital, Karolinska University hospital.

2/8: In patients with cancer, cure relies on systemic and local anti-tumor immune responses and systemic immunity vary in children with age and in relation to exposures (ex microbiome, nutrition, growth status).
