If any 🔬 peeps want a distraction from doomscrolling through the current hellscape, here is a puzzle we just uncovered:

Top row of image seems to be from the bottom of the exposed area on the camera chip, then everything shifts down

Happens full frame and with ROI. Nikon Elements, Sona sCMOs

Surprised it isn't higher, given everything

One of the hidden gems in my lab these days is a codebase to pull down our roster from the lab website and auto-gen a figure for acknowledgements slides. We use it so often.

Would this be broadly useful? LMK ... we could try to publish and support.

Is this my lab's way of telling me I should be in the office more 🤔😂

There are many great things about working at @UWGenome. This may be the greatest.

Been going for >2 years now! 🐱🗓️

Finally, a big benefit of pSABER being super bright is that you can readily image it on low NA, air immersion objectives. SABER is essentially undetectable on these systems.

We are really excited about pSABER for applications like optical screening

I was lucky enough to image another exchange-pSABER experiment carried out by Sahar targeting three RNA species on our SoRa confocal.

This might be my favorite image ever taken.

As a microscopist, it is hard to put into words just how incredible i is to see this on the scope itself.

We used DNA-exchange to label mouse minor satellite (DNA pSABER), 7SK (RNA pSABER), and the aquaporin protein (conventional IF) in FFPE sections.

The resulting images that Sahar acquired are simply gorgeous

Importantly, because pSABER simply layers more signal amplification onto SABER, we can harness all the benefits inherent to SABER as well.

E.g., we can still use DNA-exchange to multiplex

Happy #FluorescenceFriday!

Here are some HCT116 nuclei with some ... "friends" ... imaged on our SoRa confocal

Gave us quite the shock when we first saw it 😅
