And once promising stuff seems to happen clean streak some colonies, test and sequence (you can also sequence population) but clean streaked colonies is neater usually

Sounds like a good plan to keep it simple and start like that, worked often well for us. Try to see maybe based on limited intermediate OD checks (glass tubes that fit in spectrophotometer can be convenient) if strain already speeds up, so you can see soon enough if better strains take over the pop.
