Is it possible for DNA to lag a little behind if the DpnI digested product was directly loaded onto the gel? I am expecting 5.5 kb bands, these are almost 5.8 if not 6. The plasmids were at 5.5 kb before I digested them.

Both are DH5alpha cells, picked two colonies off the plate and such a stark difference in colour.

Time to head back to my gels.

Stuck in protein expression. The proteins from lane 4 onwards should be 48kDa and yet they fall between 33-43kDa. Initially I thought it must have been some contamination so I did another transformation with fresh plasmid but still the same thing.

Currently struggling with protein induction and this image came on my timeline on that app we don't talk about.

I can't be the same person who used to be scared of running a SDS-PAGE gel. Look at me now, running several gels at once.
