one more satisfying thing on the biochemical mechanisms of aberrant miRNA biogenesis in Dicer cancer hotspot settings. our friends in Shuo Gu's lab (NIH) got same results by testing a different Dicer mutation, and different species (mouse). so it must be true!! 4/4

upregulation of miRNA-3p passengers is the broadest "strand switching" effect we know of. more importantly, Dicer cancer hotspots induce loss and gain of miRNA function: important for thinking targeted therapy. congrats david jee and seungjae lee for uncovering this biochemical+genomic mystery! 3/4

Dicer cancer hotspots impair RNaseIIIb, but not RNaseIIIa activity, and was known to prevent miRNA-5p biogenesis. surprisingly, we find a selective defect in miRNA-3p levels: they directionally go UP, but only if they are normally passenger strands. we show these are functional for silencing! 2/4
