You can set up your directories on whichever drive you like, but once you have your Linux partition and directories set up, programs are only installed to those locations.

Each of those subdirectories can be mounted to whichever physical drive you like.

It makes no difference to Linux which partition on which physical drive is mounted to which directory.

So the answer is yes and no

Linux doesn't designate drives by letter. Programs are installed on whichever drive is mounted as /

Within the / directory, applications are located in /opt, and /usr

The binaries (executables) are in /usr/bin

Global configs are in /etc

Your user configs are in ~/.config/
