Finally, we asked whether FLNc dephosphorylation induces FILIP binding, which mediates its degradation. Using a pulldown assay with FILIP1 and PP1c-treated and non-treated muscle cell lysates, we found higher binding of endogenous dephosphorylated FLNc, closing the loop on this mechanism! 🔗✨ (4/5)

Could it be that a phosphatase outplays AKT and if so, which one? We studied the FLNc d20 interactome #massspec and were thrilled to identify THREE catalytic subunits of PP1! 🧬✨ In vitro assays confirmed PP1 efficiently dephosphorylates human FLNc-pS2233, even with higher activity than FastAP. Previously we discovered that FLNc is phosphorylated in its mechano-sensitive domain 20 by AKT and PKC in skeletal muscle cells. However, during mechanical stress induced by twitch EPS, we found FLNc to be strongly dephosphorylated at its AKT target site pS2234, even so AKT-pT308 was elevated 🤔(2/5)
