Inside macrophages, the particles report the onset and progression of phagosomal acidification at single-object resolution.

This work is a collaboration with B. Dumat and J.M. Mallet (ENS-PSL, Chemistry Dpt) and @florenceniedergang.bsky.social (Institut Cochin)

Each particle carries pH-responsive BODIPY dyes on its surface and a stable reference probe in the core, enabling ratiometric readouts. By decorating the membrane with Fcγ-binding ligands, we drive controlled uptake and capture the earliest steps of phagosome formation.

A collaboration between Alexandre Chargueraud, graduate student in our team (fattaccioli.github.io) and Lars Kool from the IPGG Microfabrication facility (www.plateformeipgg.fr). This work has been presented at the Micro and Nano Engineering (MNE) conference in 2024.

We also built a Python/μManager interface that automates trapping, imaging, motion control, and bubble-free priming steps. The result is a robust, fully automated platform for high-throughput single-object studies, from screening assays to mechanobiology and dynamic imaging workflows.

Confocal measurements reveal clear pressure-dependent deformation profiles, in excellent agreement with a thick-membrane model. Controlled fluidic and pneumatic actuation enables rapid, fully reversible trapping within seconds.

The issue should still be there I guess :-)
