Overall, our findings unveil a MYC-ACLY loop in which ACLY promotes MYC expression by providing the acetyl-CoA units needed for its epigenetic regulation, while MYC itself binds to the ACLY promoter and transcriptionally activates it, resulting in a feedforward loop supports T-ALL growth

Finally, using an ACLY pharmacological inhibitor, we showed consistent downregulation of MYC in human T-ALL cells , which even translated into increased survival in a human T-ALL PDX in vivo

Reverse gene set enrichment analyses using our signature upon genetic loss of MYC revealed a similarly enriched signature upon ACLY loss, suggesting ACLY could also be upstream of MYC. Indeed, luciferase reporter assays confirmed MYC regulates ACLY, and MYC loss in T-ALL led to reduced ACLY levels

Transcriptional profiling upon acute ACLY loss revealed a transcritpional programmed governed by MYC downregulation. Indeed, ACLY loss led to a drastic reduction in MYC protein levels, and epigenetic profiling showed this is likely driven by severely reduced H3K27ac levels in the MYC promoter

Interestingly, ACLY loss is completely dispensable for normal T-cell development. Next, analyses upon acute ACLY loss in T-ALL in vivo revealed a metabolic crisis with reduced acetyl-CoA levels, acumulation of AICAR and likely blocks in glycolysis, glutaminolysis and nucletotide synthesis

We first identified ACLY as downregulated upon NOTCH1 inhihibiton and overexpressed in T-ALL patients. Thus, we generated NOTCH1-induced ACLY conditional knockout primary leukemias and we observed significant antileukemic effects upon secondart ACLY loss in established leukemias in vivo
