The mechanism of STING NF-κB signaling has remained a mystery. An exciting new study from the Paludan lab now uses data throughout animal STING evolution to provide new answers – including isolating white blood cells from turtles and carp!

STING signals to NF-κB from late endolysosomal compartments using IRF3 as an adaptor - Nature Immunology
Here the authors show how the DNA-sensing cGAS–STING pathway activates NF-κB and inflammatory gene expression with delayed kinetics via post-Golgi endolysosomal signaling.

Chan Lee @harperlabhms.bsky.social reports CASM-driven V-ATPase recruitment to lysosomes for acidification upon TRPML1 activation via DMXL1-RAVE loader. With Magdalene Moran(Caraway), Sophie Helaine lab, Joao Paulo, & HarperLab teams(HMS). Funds: Warren Alpert FND/NIH www.nature.com/articles/s41...
DMXL1 promotes recruitment of V1-ATPase to lysosomes upon TRPML1 activation - Nature Structural & Molecular Biology
Lee et al. show that DMXL1, a regulator of V-ATPase assembly, is recruited to lysosomes upon TRPML1 activation in a manner dependent on conjugation of ATG8 proteins on lysosomal membranes (CASM) and p...

Brogan et al uncover a signaling pathway in which levels of the nucleotide second messenger c-di-AMP increase in response to defects in cell wall synthesis. This regulatory pathway decreases the cytoplasmic turgor pressure and protects the cell from lysis: www.nature.com/articles/s41...
