A short heated citrate buffer incubation can be used for antigen retrieval but keep it less than 15 minutes. Sodium borohydride (0.1%) incubation can also help with antigen retrieval. Happy to chat more!

If you can fix with acrolein over glut, that's going to be better for staining. An alternative is to do the staining prior following PFA fixation, fix with glut and then process for EM.

The Aves chicken one works well (Aves Lab, GFP‐1020) and I've also had success with an Invitrogen rabbit polyclonal (A11122).

What species? Are you doing immuno-EM (and immuno gold or DAB) or IHC (DAB or fluorescence)? I know a few good GFP ones depending (I'll follow up with access to my computer). There's some tricks to help restore antigenicity with glut fixed tissues but if you can fix with acrolein it's better.

I forgot about tuition. In 🇨🇦 at least, student has to pay international rate tuition for grad school, which is often very $$. The amount may change (lower) in the second+ year of MSc and post-candidacy PhD, but this is very program and university dependent.

I feel like I was maybe in grad school for a year longer than if I had transferred from my MSc to PhD, but my funding was good. (And because in 🇨🇦 many grad stipends aren't taxed, I was technically making more as a grad student than postdoc). There's pros/cons to each way and it's very individualized

I fully completed my MSc and then a PhD. For me, that was really great, as it helped me build my CV to be more competitive for awards. Depending on country/program, unlike the USA, at least in Canada, thesis based MSc usually have a stipend. I also had access to USA student loans if needed.

A lot of places will encourage an MSc or have a direct entry (with different requirements) or encourage a transfer after a year from MSc to PhD (there's usually a transfer 'exam' in addition to quals/CE). As an early participant of the 🇺🇸➡️🇨🇦 for grad school pipeline, I'm happy to chat with anyone.
