GlycoShape
@glycoshape.org
Glycan 3D database and toolbox for the reconstruction of glycoproteins from the RCSB PDB and EMBL-EBI AlphaFold repositories or own. Find us at https://glycoshape.org. Curated by head chef @elisafadda.bsky.social and sous chef @ojas-singh.bsky.social
3/3 GlycoShape is an open access library of glycan structures from MD simulations that gives users all the diff structures significantly occupied at equilibrium and tools to reconstruct the target glycoproteins with a single representative conformer (Re-Glyco) or multiple (Re-Glyco Ensemble)

2/3 Glycans aren't genetically encoded (non-templated), so they can be quite heterogeneous.

This is a problem especially in cryo-EM studies of glycoproteins, where you can assign clearly the common denominator and most rigid part of the glycan, usually the bit linked to the aglycon

Remember, if you have a structure (PDB, AF or your own) that is missing that precious little O-fucose 🔺, you can put it back were it belongs with GlycoShape Re-Glyco 👍

7/n There are many more papers to come from Silvia's work, so this is only the first part of her journey! Thank you to all the wonderful colleagues that contributed to her success thus far! 👏👏 And stay tuned for more #glycotime news! Brava Silvia!!

6/n Because of its ubiquitous presence, recognition of sialic acid is a BIG deal and evolution developed Siglecs as molecular precision tools to select specific sialic acids by recognising their biological environment [https://doi.org/10.1101/2025.06.21.660838]

5/n O-glycan sialylation can also be a discriminant for the interaction of a receptor to its target inhibiting and not favouring such interaction [https://doi.org/10.1101/2025.10.23.684103]

4/n Hypersialylation of the CD52 glycopeptide determines selection and binding to the receptor through the O-glycan and binding to Siglec-10 through the N-glycan [https://doi.org/10.1016/j.jbc.2025.108350] and [https://doi.org/10.1101/2025.06.10.658867]

3/n All of these modifications do not act the same, they are strictly context-dependent, delivering largely different messages through their largely different 3D structures and dynamics

2/n Sialic acid is a feature in many glycan and glycolipids, appearing as a terminal sugar (as a single or in multiple units) but also as a polymer within fabulous helical structures, known as polysialic acids

Also the original Sugar Drawer can be found in GlyCOSMOS glycosmos.org/glycans/grap..., you can use it to search the whole GlyTouCan library. When you use it in GlycoShape it searches for entries only in our 3D database

3/3 The interoperability between GlyCOSMOS and GlycoShape is the result of a great collaboration with @kiyokof.bsky.social lab funded by J-GlycoNet 🎉 Stay tuned for more #glycotime

1/2 Check out the page of your fav glycan in @glycosmos.bsky.social which also has all the 3D info our database[2] and a direct link to its page in GlycoShape. Makes it very easy to see how many 3D structures correspond to a single 2D structure! 😉
