What if we put all the data together?? ...Initially, vesicles are tethered at 45 nm from the plasma membrane by a ring-shaped ExHOS of 19 nm radius. As the ExHOS expands, the vesicle quickly moves towards the plasma membrane in what we called the ‘Approaching’ phase.

We next used cryoCLEM with these fluorescent temporal-rulers to quantify the vesicle movement as it approaches the plasma membrane...surprise, it is not a linear movement!

Correlating SMLM with these fluorescent temporal-rulers allowed us to model exponential kinetics controlling the conformational dynamics of a ring-shaped Exocyst Higher-Order Structure (that we named ExHOS) formed by 7 exocysts on average

Quantitative particle tracking allowed us to map protein dynamics during exocytosis and to derive fluorescent time-rulers of the process
