For #FluorescenceFriday: a very thin slice of rat kidney was labeled with 8 primary antibodies whose signal was amplified with Aluora Spatial Amplification reagents. 🔬 on the EVOS S1000 Spatial Imaging system. >400 merged fields of view at 20x.

Some hummingbirds appear colorful due to iridescence caused by the microscopic structure of their feathers. Here’s an Anna’s hummingbird feather from my yard at 460x 🧪

Under normal light fluorophore colors appear inverted: red dyes look blue, blue appear red, and green dyes are purple. Using an inverse LUT shows us this same palette on our microscopy sample. 🧪

Here's a set of three for #FluorescenceFriday. Same image, progressively higher zoom. FFPE human tonsil tissue labeled with primary antibodies against Cytokeratin, PCNA, C20, CD45RB, CD68, eCadherin, and SMA. 🧪🔬

Today in @nature.com: remarkable extended survival in pancreatic cancer patients treated with cancer vaccine.

Autogene cevumeran induces CD8+ T cells with est. lifespan of 7.7 years (range 1.5 to roughly 100 years), with 20% of clones that may outlive hosts.

These mushrooms are also the source of phalloidin - just as toxic but a useful scientific tool to label the actin cytoskeleton of cells to view under a microscope

And more recently we introduced LysoLight Green! Happy imaging and enjoy the weekend.
February 21, 2025 at 10:05 PM
LysoLight is ideally used in live cells; fixation is not recommended. Photostability is excellent, toxicity minimal, enabling long-term kinetic experiments.

Bright fluorescence is observed upon antigen-specific endosomal trafficking to the lysosome. Activation has been demonstrated with cathepsins B, D, & L. Inhibition is accomplished by blocking cathepsin, vATPase, or saturating cell-surface antigens with competing antibody.
February 21, 2025 at 10:05 PM
The LysoLight family of dyes use a unique sensor that is activated by enzymatic cleavage specific to the lysosome, and therefore reports delivery of a protein or antibody to a functional lysosome. Minimal observable background fluorescence contributes to extraordinary sensitivity.
February 21, 2025 at 10:05 PM
Several types of modern biotherapeutics require cellular uptake and lysosome delivery for efficacy. This includes ADCs, IgG-LNP, IgG-oligo, and of course extracellular targeted protein degraders like LYTACs. Existing fluorescence-based sensors report lysosomal delivery, but not catabolic activity.
