Transgenic Plodia larvae expressing the very bright mBaoJin (mStayGold variant) in its silk glands.

Sifting through new markers, we found that special UV-iridescent scales were Cluster 3:
Bab and Doublesex OFF
Abp1 and Sulfatase1 ON

Next, we wondered if given their unique morphology the UV scales cells have the signature of a distinct cell type.
The answer was a resounding YES

Single-nucleus transcriptomics recovered a few scale types including hair-like scales, regular orange or black scales, and 2 male-specific types

So how Bab is itself repressed in males to allow UV-scales?

Well, to our greatest surprise, it's not: the male state w/ Bab absent every other scale (thus allowing UV) is the default

And instead, females need the Doublesex gene to activate Bab!

Our previous study had shown that there is an developmental off switch for this : expression of the transcription factor Bric-a-Brac (bab) blocks the differentiation of UV scales

I love this SO MUCH... the plate specific neuronal driver, membrane+nuclear labelling, and the overlay with cuticle autofluorescence.... oooomph!!!
