Although we set out to discover novel regulators of MDA5, we actually stumbled on a regulator of BOTH RIG-I and MDA5! ANXA2 loss affects responses to viruses activating either or both receptors, and even activation with known RIG-I agonist 3p-hpRNA.

#WNV infection can cause #encephalopathy, and loss of ANXA2 saw poor response to virus in #iPSC #neural progenitors.
#macrophages are key sources of inflammation in response to many viruses, e.g. #flu IAV in the lungs, but iPSC-derived macrophages responded weakly in the absence of ANXA2.

Critically, when ANXA2 is absent, cells fail to induce strong innate immune responses to virus infections such as #westnilevirus #EMCV and #influenzaA. Amazingly, cells hardly "realize" they are infected with West Nile Virus if ANXA2 is absent!
