#Long-COVID, #Post-COVID und #PASC gehen häufig mit kognitiven Einschränkungen (Cog-LC) einher. Eine Studie aus Japan konnte jetzt zeigen, dass bei #Cog-LC die postsynaptische Dichte von AMPA-Rezeptoren in bestimmten Hirnregionen erhöht ist.

Long COVID primarily presents with persistent cognitive impairment (Cog-LC), imposing a substantial and lasting global burden. Even after the pandemic, there remains a critical global need for diagnostic and therapeutic strategies targeting Cog-LC. Nevertheless, the underlying neural mechanisms remain poorly understood. Given the central role of synapses in brain function, investigation of synaptic molecular changes may provide vital insights into Cog-LC pathophysiology. In this study, we used [11C]K-2 PET to characterize the density of AMPA receptors (AMPARs) on the post-synaptic cell surface, which are crucial synaptic components in brain signalling. Statistical parametrical mapping was used to spatially normalize and apply independent t-test for a voxel-based comparison. We selected patients with Cog-LC (n = 30) based on Repeatable Battery for the Assessment of Neuropsychological Status assessed persistent cognitive impairment and healthy controls (n = 80) with no diagnosed neuropsychiatric disorders. The primary objective was to compare [11C]K-2 standardized uptake value ratio with white matter (SUVRWM) as a reference region between patients with Cog-LC and healthy controls, and to define the regional extent of differences. The secondary objective was to examine associations between [11C]K-2 SUVRWM and plasma concentrations of cytokines or chemokines. As an exploratory objective, we tested whether [11C]K-2 PET data could distinguish Cog-LC from healthy controls using a partial least squares based classification algorithm. A voxel-based comparison (P < 0.05, T > 1.66, one-tailed, false discovery rate control) and a volume of interests analysis (P < 0.05, Bonferroni multiple comparison) demonstrated that increased index of AMPAR density in large parts of the brains of patients with Cog-LC compared with that in healthy controls…

In 1979, Apple introduced Apple Pascal along with the Apple Pascal Operating System for the Apple II. It paved the way for later Apple OSes, including SOS, #Lisa_OS, and #Mac_OS (which were written in #Pascal as well). Choosing Pascal laid the foundation for the high stability of Apple's OSes.

Bild der frisch geputzten Stolpersteine von Siegmund, Hanny und Clara Cohen, die von den Nazis verfolgt, gedemütigt, entrechtet und schließlich ermordet wurden. Das Manufaktur- und Bekleidungsgeschäft der Familie wurde in der Reichspogromnacht 1938 verwüstet und Siegmund Cohen von der SA so schwer zusammengeschlagen, dass er kurze Zeit später an den Folgen starb. Ein Arzt und später eine ordentliche Bestattung blieben ihm verwehrt. Clara und Hanny Cohen mussten 1940 in ein Judenhaus umziehen. Hanny heiratete dort Bernhard Meyer. Nur drei Monate später wurden alle drei mit 570 weiteren Personen aus dem Umkreis in das Ghetto Minsk deportiert. Clara Cohen verstarb wahrscheinlich schon im Winter 1941. Wer bis zum Som­mer 1942 über­leb­te hat­te, fand schließ­lich sei­nen Tod am 28. und 29. Juli im Gas­wa­gen oder durch die Er­schie­ßun­gen an den Gru­ben in Maly Tros­te­nez im Sü­den von Minsk. Das genaue Todesdatum aller drei ist unbekannt. Hinter den Steinen sitzt ein kleiner Bär, Kurt, auf einem pinken Lappen und spiegelt sich im glänzenden Metall der Stolpersteine. Er hält ein rotes Herz und ein Schild mit einem Zitat des Journalisten Mario Sixtus. Es lautet: "Dass wir heute im Stadtbild so viele Stolpersteine haben, liegt übrigens daran, dass sich mal eine Regierung an bestimmten Menschen im deutschen Stadtbild störte. (Mario Sixtus)"

Apple's legendary #Pascal syntax poster from 1979. It was created by Jef Raskin to adapt the syntax of Bill Atkinson’s #Apple_Pascal compiler. The poster was over the desk of every programmer at #Apple and over the desks of many programmers outside of Apple.

Gestational #diabetes mellitus (GDM) is linked to significant adverse outcomes for both mothers and their offspring. Since #SPINA-GBeta outperforms #HOMA-Beta, its use is advised as a preferred calculated #biomarker for the evolution and pathophysiology of #GDM.

Molecular Mechanisms in gestational diabetes mellitus (GDM). Pregnancy-induced insulin resistance is driven by placental hormones (human placental lactogen (hPL), placental growth hormone, estrogen, progesterone, cortisol) and inflammatory cytokines (tumour necrosis factor α (TNF α), interleukin 6 (IL-6), interleukin 1β (IL-1β)). Inadequate β-cell adaptation due to mitochondrial dysfunction, and impaired incretin signaling leads to hyperglycemia. Dysregulated adipokines (decreased adiponectin, increased leptin, resistin, visfatin), oxidative stress, and epigenetic modifications (deoxyribonucleic acid (DNA) methylation, non-coding ribonucleic acids (RNAs)) further exacerbate disease and contribute to adverse maternal and fetal outcomes.

There is an ongoing debate in #endocrinology about whether to treat #hypothyroidism with L-T4 alone or a combination therapy with L-T4 and L-T3. But what about patients’ preferences? The majority would choose the combination therapy, as shown in a meta-analysis.
