WTH… is VPS35 the next candidate to fight mitochondrial dysfunction? I just leave it here…

Not only we recover mtDNA damage! Look all these transcripts!

We then used Drosophila and expressed VPS35…. 💥 💥 🤯

Ok! But let’s be serious… we noticed that overexpression of the Retromer increased mtDNA release… could we use this to clean up mtDNA damage?

Not only that! Whenever mtDNA is released, look who’s there?! A lysosome

What then? We found that the leak gets inhibited upon inhibition of BAX1 but not VDAC1

Surprise! No, mtDNA leaks independent from MDVs

What about the mtDNA? Is it also release through MDVs?

We took a deep look at the Retromer! This guy was always around! We found that mtDNA stress triggers the formation of MDVs, but wait… they were lysosomal dependent… interesting…

Who was there? Vesicle proteins! Lysosomes! Mitochondria matrix proteins! How?

We started with SplitTurboID between mitochondria and endosomes to find out the proximity proteome upon mtDNA stress…
