Critically, the final engineered cells are fully functional: they evade NK killing & establish T-cell tolerance.
This work provides a platform for programming complex human cells, from immune design to epigenetic rules.

This allowed us to ask: what controls this epigenetic ghost? By comparing a "genes-only" locus to one with native intergenic DNA, we found the answer.

This non-coding DNA contains the "software" that actively promotes silencing and resolves the 👻. 🧵

We built REWRITE to tackle a huge problem in cell therapy: HLA mismatch. We deleted massive regions and wrote in 100 kb synthetic haplotypes.

But this led to a surprise: the engineering left a heritable "epigenetic ghost" of active expression. 🧵

Got any good ideas of what, other than lawsuits and protests? You're a leader in the community after all. I'm at a loss for what to do, especially from liberal NYC.
