Although TFs are difficult to drug, several attempts have had success (i.e AR & Myc). Future efforts to specifically target TFs may involve advances in targeted protein degradation (oligoTRAFTACs). This could facilitate 'modular' approaches to tailor treatments to specific TFs.

Our understanding of which lineage-specific TFs drive cell fate has led to intriguing discoveries in functional genomics studies in the cancer field (thanks to large scale efforts like DepMap). These TFs represent some of the most specific and potent genetic dependencies in distinct cancers.

Starting with core regulatory circuit we review the importance of these networks in cell fate decisions and controlling regulatory elements like enhancers.
