Collectively, these studies highlight the complex contribution of membrane estrogen signaling to the hypothalamic regulation of reproduction.

Therefore, E4 seems to act as a weak estrogen agonist on this system. This contrasts with its antagonizing action on the circuit underlying the estrogen-induced positive feedback (www.eneuro.org/content/11/1...).

Estetrol (E4), a natural estrogen which antagonizes membrane ERα activation in some cell types mimicked the effect of E2 on the negative feedback and its cellular correlates.
March 31, 2025 at 11:55 AM
Treatment with a higher dose of E2 alleviates the LH dysregulation and some of the associated hypothalamic measures.
March 31, 2025 at 11:55 AM
This absence of negative feedback is paralleled by a lack of changes of different hypothalamic markers of this process (Kp, PR and NK3R).

Study shows that C451A-ERα mice, which lack mERα signaling, respond normally to ovariectomy (rise in LH) but do not respond to chronic estrogen exposure as wildtype.

Congrats!
