The nucleus is fantastic, but can it beat #cilia and #flagella 😉 here stained for tubulin (magenta) with an intraflagellar transport train (green) after #expansion microscopy and #STED or shown by classic TEM. JCS @jcellsci.bsky.social found the solution and is featuring both! 😍

Thanks to a new mass spec pipeline for analysis of the #tubulin tail, the team of Julia Chamot-Rooke and Mariette Matondo revealed a few surprises for #trypanosome beta-tubulin:
-a new glutamylation site
-far less exhaustive detyrosination

I forgot to say that the tagged tubulin incorporates very well in the spindle, here after a 4-hour induction (green, magenta is axoneme + transition zone). However, we could not see a defined polarity, in contrast to flagellar or cortical microtubules.

We propose a model where the new flagellum grows in a linear manner while the old one is locked but shrinks slightly, hence requiring unlocking and compensation after cell division, leading to such multicoloured flagella following induction/deinduction/reinduction of tagged tubulin 😉
