Thyroid hormone receptor β1 (TRβ) downregulation correlates with poor prognosis in clear cell renal cell carcinoma (ccRCC). Restoration of TRβ and the peroxisome proliferator-activated receptor gamma coactivator 1-alpha suppresses tumor progression in ccRCC through Uncoupling protein 1 mediated tumor cell “slimming” and optic atrophy 1/mitofusin 2 dependent mitochondrial fusion. Abstract The abnormal accumulation of lipids is a hallmark of clear cell renal cell carcinoma (ccRCC). Both the thyroid hormone receptor β1 (TRβ) and peroxisome proliferator-activated receptor gamma coactivator 1-alpha (PGC1α) are key regulators of mitochondrial function and lipid metabolism. However, their specific interaction and influence on ccRCC development and lipid accumulation remain poorly understood. This study identified genes jointly regulated by TRβ and PGC1α, which are implicated in lipid browning and mitochondrial fusion. Mechanistically, T3-activated TRβ interacts with PGC1α to transcriptionally upregulate PGC1α, UCP1, and mitochondrial fusion genes OPA1 and MFN2, thereby enhancing mitochondrial activity, promoting lipid utilization, and suppressing ccRCC progression. These results indicate that the mitochondrial and metabolic effects of TRβ in ccRCC are mediated through PGC1α expression and function. Activation of the TRβ/PGC1α through hormonal and pharmacological means may offer a promising therapeutic approach for ccRCC.

The normal range of TSH is 0.4-5.0. I am at .8 right now, but if it’s above 1.5 I do better with regulating my body temperature. I have thyroid nodules that stay stable, but I haven’t had a dose increase in years. I think he will increase it.

Then we can talk about my crappy bone density.

Interesting, I've long suspected thyroid issues for me but I never test out of range. I'm always at the low end of normal, yet I have all the symptoms of thyroid disregulation and literally everyone in my family has hypothyroidism and is on Synthroid

It really does. Tomorrow I see my endocrinologist and I’m going to ask for an increase in my thyroid meds, there is a sweet spot with those levels that lessens the attacks for me. I’m testing normal but the low end of normal right now.

Dr. Susan Baumgaertel at Seattle's Polyclinic Madison dismissed my thyroid cancer as menopause for 2+ yrs--she never even sent me to an endo--& it was Stage 3 when I ended up in the ER.

Until the ACA I was uninsurable due to pre-existing conditions.

Every time I changed jobs there was a 90 day period that my thyroid meds and MD visits weren't covered. I haven't had a thyroid since 1992.

The ACA changed all of that.
