SPOCQ has the great advantage over SPAAC as the proteinogenic amino acid tyrosine offers direct access to a rapid and clean method for protein modification, especially to convert therapeutic antibodies into antibody-drug conjugates (ADCs) and bispecific T cell engagers (BiTEs) (vide infra).

We used a stable quinone and a range of strained cyclic alkenes and alkynes such as BCN and DBCO for in-depth kinetic studies of the SPOCQ click reaction (which is typically >100 times faster than the more commonly used SPAAC).

With deltaH++ of <2 kcal/mol, these cycloadditions are entropy-driven. This has been mentioned before in the literature, but now we have the exact numbers, obtained by stopped-flow spectroscopic analysis!

Reaction rates in SPAAC but not in SPOCQ correlated to sp-sp3 bond angles!
