Retired substance abuse counselor here. I have always doubted Kennedy’s alleged recovery. Majority of opioid addicts relapse across the lifespan unless treated with methadone, suboxone, or naltrexone. Cabinet nominees should submit to drug testing.

Wow. I wouldn't be surprised if tirzepatide holds its own against naltrexone.

My experience on sema is actually like a mild form of how I've heard naltrexone makes you feel... For me it's all the downsides of booze but very little of the upside/buzz.

Dysfunctional Transient Receptor Potential Melastatin 3 (TRPM3) ion channels are associated with the pathophysiology of long COVID due to reduced calcium (Ca2+) influx, negatively impacting cellular processes in diverse systems. Accumulating evidence suggests the potential therapeutic benefits of low-dose naltrexone (LDN) for people suffering from long COVID. Our study aimed to investigate the efficacy of LDN in restoring TRPM3 ion channel function in natural killer (NK) cells from long COVID patients. Methods: NK cells were isolated from nine individuals with long COVID, nine healthy controls, and nine individuals with long COVID who were administered LDN (3–4.5 mg/day). Electrophysiological experiments were conducted to assess TRPM3 ion channel functions modulated by pregnenolone sulfate (PregS) and ononetin. Results: The findings from this current research are the first to demonstrate that long COVID patients treated with LDN have restored TRPM3 ion channel function and validate previous reports of TRPM3 ion channel dysfunction in NK cells from individuals with long COVID not on treatment. There was no significant difference in TRPM3 currents between long COVID patients treated with LDN and healthy controls (HC), in either PregS-induced current amplitude (p > 0.9999) or resistance to ononetin (p > 0.9999). Discussion: Overall, our findings support LDN as a potentially beneficial treatment for long COVID patients by restoring TRPM3 ion channel function and reestablishing adequate Ca2+ influx necessary for homeostatic cellular processes.

ID: Long COVID research updates: A small mechanistic study analyzed T cells and chronic inflammation in COVID-19. A recent paper found that low-dose rapamycin alleviated symptoms of fatigue and PEM in people with ME. A phase 2 clinical trial is recruiting in Boston to test Pyridostigmine (Mestinon) and Low-Dose Naltrexone (LDN) for ME. A small mechanistic study in Frontiers in Immunology analyzed chronic inflammation in COVID-19. Researchers assessed over 70,000 blood cells from eight participants across different disease states. They honed in on issues with a type of white blood cell, called memory CD8+ T cells, that respond to specific pathogens and may remain activated beyond the acute phase of COVID-19, leading to an impaired immune response. The researchers also identified seven genes that may help predict immune dysfunction. While this analysis focused on people with acute COVID-19, the study supports growing evidence about T-cell dysregulation in Long COVID. A recent paper found that low-dose rapamycin alleviated symptoms of fatigue and post-exertional malaise in people with myalgic encephalomyelitis (ME). The small study included 86 participants — all taking the drug, with no placebo arm — of whom 40 completed the full 90 days, taking 6 milligrams of the drug per week. Researchers measured specific markers of the metabolic system as outcome measures and found improvements in both that may link to potential biomarkers. “These findings support the safety of once-weekly low-dose rapamycin in [ME] and justify further investigation in larger controlled trials,” the authors wrote. A phase 2 clinical trial is recruiting in Boston, Massachusetts to test the drugs Pyridostigmine (Mestinon) and Low-Dose Naltrexone (LDN) for ME. Pyridostigmine is a drug currently approved for the autoimmune disease myasthenia gravis. LDN is being studied in Long COVID and ME (higher doses of naltrexone are used to treat addiction). 160 participants will be divided into four groups: Pyridostigmine/LDN, Pyridostigmine/placebo, LDN/placebo, placebo/placebo. On what did work for me:

• CBT (cognitive behavioral therapy, you perverts) that I ironically learned not from anyone in a recovery setting, but rather from a failed attempt to quit smoking

• MAT (medication assisted treatment). Naltrexone to be exact. Which was mentioned as an aside for IOP plant

I carried a stack of text books up 2 flights of stairs today and it was only uncomfortable! I even cleaned and scrubbed dishes and countertops and the stove top!!! Yippeeeeeeee

My chronic illness is significantly more tolerable since getting on low-dose naltrexone. If I skip it for even a day or two, the full-body pain comes roaring back, and I think, “I can’t believe I used to live this way every day.” But of course, I only lived that way because I thought I had to. 💙

In one recent study, O’Malley found naltrexone to be effective in limiting consumption among college-age drinkers. The drug helped subjects keep from going over the legal threshold for intoxication, a blood alcohol content of 0.08 percent.
