How does this change in GVHD? We know that donor T cells attack the recipient, but is this the whole story? No! CD8s were only enriched in some patients. We suggest a major role for plasma cells, macs and neutrophils, which correlate with worse clinical manifestation.

What do these represent? Lots of known unconventional T cell subsets (MAITs, NKTs, various thymic developmental subsets, KIR+ CD8s, and Tregs) and lots of unknown discrete unconventional populations.

Intuitively I associate nucleic acid-sensing TLRs with CD8s because antiviral + type 1 IFN induction
but tbh I think it's probably much more about tropism so that you can get antigen into cytosol so cross-presentation would be key there so anything that gets into cDC1s better will probably be best

In short, BB700 was the big winner. It continuously labeled for over 6 hours in vivo (crazy right???). Applying it to a transplantable tumor model, we found we could identify roughly 7-fold more tumor specific CD8s that had recently migrated compared to IV administration. 9/n

GPT tells me the boosters are sAgs and ignite effector CD8s (and CD4s). This is done non-specifically, so they immediately emit cytotoxic granules and cytokines non-specifically. Then they die or are exhausted and have to be replenished from a limited pool of naive progenitors.

Wow, I would have expected better QC from the Nussenzweigs! So weird! Did you read that piece in immunity where hyperreactive B cells appear to induce CD8s into killing them? I haven't read it fully yet, but was thinking of sending it your way since something similar happens in GCs but via NK cells.

That’s a very challenging expt to do bec it’d be difficult to discern antiviral effects of endogenous vs donor exhausted T cells. But prior studies show that depleting CD8s during chronic SIV infection leads to increase in viral loads. So even if CD8s appear exh, they still retain some antiviral fx.
