Fair enough, but I don't think it's much of a stretch to suggest that there exist differences in cultured and live preps. By protein trafficking, I meant in membrane structures (like channels) which *seem* to be different in cultured cells, from what I've gleaned.

That's all fine and well but the translatability cannot be assumed 1:1 for a ton of reasons

It's known that, at least in neuronal cell types, heterogeneity of channels in cultured cells is drastically different from native cells. For example, cultured human-derived cardiac myocytes require strong hyperpolarizing current to elicit APs, which you don't see in an in vivo prep.

the danger with this is that protein expression/trafficking in culture is just not the same as an intact organismal system. See- electrophysiology studies in cultured neurons. It's cool, but I would still prefer an animal model and human trials over cells in wells
