When my teacher told me that there existed machines that could do automated Sanger sequencing, I knew that this was exactly what I wanted to do...
The problem is that pacbio is super expensive. The enrichment probes for chrY regions aren't selective enough to specifically distinguish regions you prefer, and you can only enrich for known sequences. So WGS is still the best for long reads.
